In vitro Regeneration of the 'Piel de Sapo' and 'Cantaloupe' Melon

Melon crop has relevant socioeconomic importance, especially in the Northeast region of Brazil. In melon breeding programs that use biotechnological techniques, it is essencial to develop efficient in vitro plant regeneration methods. The objective of this work was to develop a regeneration protocol from melon cotyledonary explants. The experimental design was completely randomized, in a factorial scheme (2 x 5), two melon cultivars (‘Pele de Sapo’ and ‘Cantaloupe’) x five concentrations of BAP (0, 4.44, 8.88; 13.32 and 17.75 μM), with five replicates of five test tubes containing one explant each. Callus and bud formation percentages/cotyledon, number of buds formed/callus and callus fresh mass were evaluated at 35 days after explant in vitro inoculation. The data were submitted to analysis of variance and the means were compared by the Tukey test between cultivars, and by regression analysis for doses of BAP. In culture medium without BAP there was no bud formation being necessary to add it in both cultivars. The number of buds regenerated per explant was higher in 'Cantaloupe'. In 'Cantaloupe', 68% of calli formed buds, in the culture medium with 8.88 μM BAP, and in 'Pele de Sapo', only 12%. Cantaloupe presented the highest fresh masses callus and number of buds regenerated per explant, standing out that it is more in vitro responsive than Pele de Sapo.
